After the release of the teen comedy Superbad, the film earned huge achievements in the box office. Many critics hailed Christopher Mintz-Plasse's performance as McLovin. For the role, he got a nomination for the MTV Movie Award for Best Breakthrough Performance. Mintz-Plasse next appeared with Paul Rudd and Seann William Scott in 2008 David Wain's film Role Models as Augie Farques, a geeky adolescent fixated on medieval role-playing.

He got his next big role when he appeared in Kick-Ass as the hero Red Mist, which was intended to be a minor role until director Matthew Vaughn saw Mintz-Plasse tryout and was impressed with his acting abilities. He guest featured in a May 2010 episode of the comedy Party Down as Kent, a companion of the character Roman. Mintz-Plasse provided his voice in the film Marmaduke, as the adapted pooch Giuseppe. The same year Mintz-Plasse was featured in Kid Cudi's Erase Me music video.

Christopher had a little role as Tommy in the film Pitch Perfect. He was supposed to assume the lead role in the CBS sitcom Friend Me and even taped a few episodes, however the series was dropped before it could air after series maker Alan Kirschenbaum tragically committed suicide. As a promotion for the computer game Far Cry 3, Mintz-Plasse featured in The Far Cry Experience, a series of online ads, depicting an anecdotal variant of himself caught on an island constrained by a psychopathic privateer named Vaas. Mintz-Plasse repeated his role of Chris D'Amico in the sequel Kick-Ass 2, which was released in August 2013. He saw the role as darker than some other he had played to date and contracted an acting coach to assist him with the more serious scenes.

In January 2020, it was announced that the film, How to Train Your Dragon: The Hidden World was added to the Oscar nominations list for the Best Animated Feature category. Mintz-Plasse has a supporting role in the film as the voice of Fishlegs.
